Oxidation of Isodrimeninol with PCC Yields Drimane Derivatives with Activity against Candida Yeast by Inhibition of Lanosterol 14-Alpha Demethylase

Isodrimeninol is a natural product isolated from the bark of the tree Drimys winteri. It was used for the synthesis of four other sesquiterpenoids through oxidation with PCC. The products were tested for their antifungal activity against various Candida species that cause Candidiasis, an opportunistic yeast infection responsible for more than 50000 deaths per year worldwide. Isodrimeninol and one of its oxidation products bind to the catalytic site of lanosterol-14-alpha-demethylase as shown by in-silico analysis, which provides the mechanistic rationale for the observed antifungal activity.
