Polyelektrolyte: Synthese, Charakterisierung und Komplexbildung
Polyampholyte
Neue, streng alternierende Polyampholyte werden durch radikalische Polymerisation von N,N-Diallyl-N,N-dimethylammoniumchlorid mit N-substituierten Maleinsäure-amiden synthetisiert. Als Seitenketten werden dabei Alkyl- bzw. Arylreste unterschiedlicher Hydrophilie bzw. Hydrophobie eingefügt:

Durch Variation des pH-Wertes gelingt es dabei die Konformation der Polyampholyte von einer mehr gestreckten Konformation (als Polykation bzw. Polyanion) in eine mehr geknäuelte Konformation (als Polyampholyt am isoelektrischen Punkt) zu überführen [3].

Polyplexbildung
Die Komplexbildung zwischen dem Polyanion DNA und kationischen Polyelektrolyten wird als Polyplexbildung bezeichnet und ist von besonderem Interesse für den Gentransfer. Dabei hat sich Polyethylenimin (PEI) in Bezug auf die Transfereigenschaften als sogenannter "Gold"-Standard erwiesen. Der Nachteil des verzweigten PEI ist jedoch seine Zytotoxizität. Basierend auf diesem Erkenntnisstand haben wir in Kooperation mit dem Leibniz-Institut (Dresden) das Komplexbildungsverhalten von zuckermodifizierten PEI-Proben und PEI-umhüllten Goldnanopartikeln untersucht.
